报告题目:Optimization and Applications of CRISPR/Cas System in MammalianCells

摘要

In life science researches, deletion, insertionor modification of DNA sequence can help us find gene function and regulatoryelements. In recent years, CRISPR/Cas system start to be used as a simple andhigh-efficiency genome editing tool. We developed a focused CRISPR/Cas-based lentivirallibrary in human cells and a method of gene identification based on functionalscreening and high-throughput sequencing analysis. Using knockout libraryscreens, we successfully identified the host genes essential for theintoxication of cells by anthrax and diphtheria toxins, which were confirmed byfunctional validation. To improve the efficiency of producing reliable genomicmodifications, we designed a method using a linear donor fragment containing areporter system. Combined with a homologous recombination-independent knock-instrategy, we successfully enriched those cell clones that specifically carrythe target gene mutations. We also developed a CRISPR/Cas9 imaging system forpainting an entire chromosome in a live HeLa cell, making it possible for thefirst time to visualize the dynamics of a particular chromosome through thecell cycle.

 

简历:

        周悦欣,北京大学生命科学学院博士,主要研究方向为基因编辑技术的开发和应用。迄今为止,周悦欣博士已经在《Nature Nature Biotechnology Cell Research》等顶级期刊发表了论文7篇,总引用超过200多次,同时申请有多项国家专利,其中一项专利获得18届中国专利优秀奖。 同时,周博士也获得了博士研究生国家奖学金,北京大学三好学生标兵,北京大学研究生学术十杰等荣誉.。
